You can arrange it with Andina del Sud which is nearby the central tourist kiosk. They have 1-day tours to Puerto Varas, Parque Nacional V Perez Rosales, Petrohue, Lago Todos los Santos, Peulla and return. They charge $37, meals included (or $27 without) but it's cheaper if you buy this tour from the kiosks inside the bus terminal.
